
The authors presented the development and application of a multi-PCR for rapid detecting of typing dengue viruses in hemorrhagic patients. The primers were designed to anneal specifically to any of the four dengue virus types. First, primers D1 and D2 were applied on RT-PCR to produce a specific DNA of dengue virus with 511 bp in size. The DNA were used to type dengue virus by specific primers in a second round of PCR amplication. Yielding DNA products the unique sizes of which were diagnostic for each dengue virus serotypes. Initially, the application of m-PCR in 80 serum amples of patients with hemorrhagic fever in outbreak in 2009 in Hanoi. Results showed that 36 samples (45 percent) were positive with DENV and all of them were serotype I. The results showed that m-PCR method can be widely applied in the early detection of dengue virus serotype in hemorrhagic patients.
