Background: The aim of this study was to investigate genetic variations of STAT4 gene and TRAFI-C5 locus in relation to susceptibility to and the severity of IgAN patients. Materials and methods: This study was performed in Karolinska Institutet, Sweden. Two hundred and twelve unrelated patients with biopsy-proven IgAN and 477 healthy were included in the study. Genotyping for single nucleotide polymorphisms (SNPs) of the STAT 4 gene and TRAF1-C5 locus was performed in all samples using a TaqMan allelic discrimination assay. Results: The genotype frequencies and allelic frequencies of rsl0181656 of STAT4 gene and rs3761847 in the TRAF1-C5 locus were no significant difference between control group and the IgA nephropathy patient group as well as IgA nephropathy patients groups with or without progression. Conclusions: the data showed that there was no an association of two single nucleotide polymorphisms (SNPs) belonging the STAT 4 gene and TRAF1-C5 locus with the susceptibility to and the severity of IgAN nephropathy.
